I would go DC to Skyline Drive (Front Royal VA). Then hit Charlottesville VA (where UVA is located). Charlottesville VA to Asheville NC (and/or Boone NC). Then back to DC.

Just as an update - we have booked DuPont Yurts for two nights staying at various places on the way down (including Damascus for the Creeper Trail). We have split the drive back from DuPont with an overnight stay near Roanake (so will work out about 4 hours each day).
Really looking forward to it.
Need to get in touch with some hire companies. Think we will do creeper trail as a family but will also get some opportunities to go out on my own (or with my son) whilst my wife sits relaxes and reads!

Just wanted to say thanks for the recommendations.
We had a fantastic time.
We got an AirBNB in the centre of Damascus. My son and I did the creeper trial. My wife went to a diner got pancakes and read. We all really like Damascus and Abingdon.
We also spent 2 nights at DuPont yurts which was fantastic. The Yurts are lovely and in a beautiful area. We got ice creams from the kanteen and did a jeep tour with Asheville Jeep Tours which included some hiking in the Pisgah forest.
We drove some of the Blue Ridge highway as well.
A great area of the country. Everyone was friendly and welcoming.
We are already planning next year and thinking about Colorado but on a slightly slower time pace (so 4 or so nights in one place). There will be more time there for riding so look out for a thread looking for more reccomendations!
